研究未讨论温度对探针回收率的影响。(2)流速可直接影响待测物质的回收率,探针回收率可随流速增大而减小。体内微透析实验要求在一定的时间内获得足够定量测量的样品容积,经对不同流速(0.5,1,2,3,4 μL/min)体外回收率的考察,流速在2 μL/min时符合上述要185 求,且增量法和减量法测得的探针回收率相一致,由此证明体内反透析法(减量法)测得的探针回收率可用于体内透析(增量法)物质浓度的计算。 通过对微透析探针回收率的研究,确定了反透析法可作为钩藤碱体内回收率的测定方法,回收率的测定是计算体内药物浓度的基础。实验结果还显示不同探针间回收率存在一定的差别,因此,每次探针植入后都应测定回收率用于本次体内药物浓度的计算。本实验还对190 灌流速度、采样量及采样时间等微透析参数进行了初步的研究,最终选择流速为2 μL/min,采样时间为15 min,样品量为30 μL作为体内微透析参数设置的参考值。 5.结论 微透析技术可用于钩藤碱的体内药动学研究,反透析法可作为体内研究中钩藤碱探针回收率的测定方法。 195 [参考文献] (References) [1] E.
